About Teriflunomide Function
Teriflunomide is an active metabolite of leflunomide and is a drug used to treat rheumatoid arthritis. It inhibits pyrimidine synthesis and is therefore effective in reducing T-cell and B-cell proliferation.
Teriflunomide acts primarily as an inhibitor of dihydroorotate dehydrogenase (DHODH), a key mitochondrial enzyme involved in the de novo synthesis of pyrimidines in rapidly proliferating cells. By reducing the activity of high-affinity proliferating T and B lymphocytes, teriflunomide may attenuate the inflammatory response to autoantigens in MS. Therefore, teriflunomide can be considered a cytotoxic drug for leukocytes rather than a cytotoxic drug.
Teriflunomide is indicated for patients with relapsing forms of MS, helping to reduce the frequency of relapses and slow the progression of disability.
